A pesar de d\u00e9cadas de investigaci\u00f3n, la comprensi\u00f3n de su inicio y progresi\u00f3n sigue siendo incierta, lo que complica el diagn\u00f3stico y tratamiento temprano. Los avances recientes en la fisiopatolog\u00eda de la EP sugieren tratamientos prometedores para frenar la progresi\u00f3n de la enfermedad, pero revertir la degeneraci\u00f3n celular sigue siendo inalcanzable. Con la aparici\u00f3n de nuevas terapias, la necesidad de herramientas de detecci\u00f3n temprana es urgente. Sin embargo, faltan biomarcadores validados para el diagn\u00f3stico de la EP, que actualmente depende de escalas subjetivas como Hoehn y Yahr o de t\u00e9cnicas de imagen m\u00e9dica costosas. La acumulaci\u00f3n de prote\u00ednas \u03b1-sinucle\u00edna (\u03b1-Syn) mal plegadas en la patolog\u00eda del p\u00e1rkinson ha despertado inter\u00e9s, pero su papel diagn\u00f3stico a\u00fan requiere m\u00e1s investigaci\u00f3n. Hallazgos recientes de \u03b1-Syn en ves\u00edculas extracelulares derivadas de neuronas (NDEV) de pacientes con EP sugieren un potencial para nuevos m\u00e9todos de diagn\u00f3stico. Nuestro proyecto propuesto, V\u03b1MPiRE, tiene como objetivo llevar a cabo un estudio longitudinal con 600 participantes con EP y 600 sin EP, utilizando una metodolog\u00eda de casos y controles ajustada por cl\u00fasteres, para explorar las isoformas de \u03b1-Syn y biomarcadores relacionados en NDEV para la detecci\u00f3n temprana de la EP.<\/p><\/div><\/div>\n<\/div>\n<\/div><\/div>\n\n\n\n<div class=\"wp-block-uagb-container uagb-block-db273ceb alignfull uagb-is-root-container\"><div class=\"uagb-container-inner-blocks-wrap\">\n<div class=\"wp-block-uagb-container uagb-block-4de41fc2\">\n<div class=\"wp-block-uagb-info-box uagb-block-93a25b56 uagb-infobox__content-wrap  uagb-infobox-icon-above-title uagb-infobox-image-valign-top\"><div class=\"uagb-ifb-content\"><div class=\"uagb-ifb-title-wrap\"><\/div><p class=\"uagb-ifb-desc\">Planeamos desarrollar y validar una innovadora prueba de diagn\u00f3stico in vitro (IVD) capaz de detectar las etapas m\u00e1s tempranas del p\u00e1rkinson y estimar el pron\u00f3stico y la progresi\u00f3n de la enfermedad. Utilizando modelos de IA para generar algoritmos de an\u00e1lisis de datos y en colaboraci\u00f3n con laboratorios anal\u00edticos l\u00edderes y fabricantes de IVD, nuestro objetivo es garantizar la fiabilidad y viabilidad del prototipo desarrollado. A trav\u00e9s del trabajo del consorcio, prevemos licenciar la propiedad intelectual generada para impulsar la comercializaci\u00f3n de nuestros resultados.&nbsp;<br>Se realizar\u00e1n dos extracciones de muestras de sangre con un intervalo de 24 meses a los participantes con p\u00e1rkinson, y una \u00fanica extracci\u00f3n basal a los controles sin p\u00e1rkinson. Todos los participantes ser\u00e1n seguidos de forma regular durante este per\u00edodo de 24 meses para monitorizar la evoluci\u00f3n de la enfermedad y el tratamiento. Aquellos controles sin p\u00e1rkinson que desarrollen la enfermedad formar\u00e1n parte de una tercera cohorte (se esperan unos 24 sujetos, seg\u00fan una incidencia del 4%) que servir\u00e1 para confirmar la sensibilidad del test en sujetos asintom\u00e1ticos.\nEl aspecto \u00fanico del proyecto es que anticipamos poder detectar ese 4% de participantes sin p\u00e1rkinson que desarrollar\u00e1n la enfermedad, demostrando as\u00ed el valor de estos biomarcadores para identificar el p\u00e1rkinson en etapas tempranas.<\/p><\/div><\/div>\n<\/div>\n\n\n\n<div class=\"wp-block-uagb-container uagb-block-b19e2dd2\">\n<div class=\"wp-block-uagb-info-box uagb-block-8bbb62ad uagb-infobox__content-wrap  uagb-infobox-icon-above-title uagb-infobox-image-valign-top\"><div class=\"uagb-ifb-content\"><div class=\"uagb-ifb-title-wrap\"><p class=\"uagb-ifb-title-prefix\">El plan<\/p><h2 class=\"uagb-ifb-title\">Desarrollo de una prueba IVD de vanguardia para la detecci\u00f3n temprana y el pron\u00f3stico del p\u00e1rkinson<\/h2><\/div><\/div><\/div>\n<\/div>\n<\/div><\/div>\n\n\n\n<div class=\"wp-block-uagb-container uagb-block-f48efe46 alignfull uagb-is-root-container\"><div class=\"uagb-container-inner-blocks-wrap\">\n<div class=\"wp-block-uagb-container uagb-block-aff9a926\">\n<div class=\"wp-block-uagb-info-box uagb-block-ddda4ef9 uagb-infobox__content-wrap  uagb-infobox-icon-above-title uagb-infobox-image-valign-top\"><div class=\"uagb-ifb-content\"><div class=\"uagb-ifb-title-wrap\"><p class=\"uagb-ifb-title-prefix\">El resultado<\/p><h2 class=\"uagb-ifb-title\">Validando una prueba IVD revolucionaria para transformar la detecci\u00f3n y el manejo del p\u00e1rkinson<\/h2><\/div><\/div><\/div>\n<\/div>\n\n\n\n<div class=\"wp-block-uagb-container uagb-block-7493c854\">\n<div class=\"wp-block-uagb-info-box uagb-block-4b4cc374 uagb-infobox__content-wrap  uagb-infobox-icon-above-title uagb-infobox-image-valign-top\"><div class=\"uagb-ifb-content\"><div class=\"uagb-ifb-title-wrap\"><\/div><p class=\"uagb-ifb-desc\"><br>El prototipo ser\u00e1 validado por su capacidad discriminativa, utilizando el primer conjunto basal de muestras de pacientes con p\u00e1rkinson y controles sin p\u00e1rkinson, as\u00ed como por su habilidad para detectar la progresi\u00f3n del p\u00e1rkinson comparando los datos basales con los de los 24 meses y las muestras de sangre.<br>Una mejora en la detecci\u00f3n temprana podr\u00eda permitir diagnosticar antes 270,000 nuevos casos de p\u00e1rkinson, mejorar el manejo de la enfermedad de 9.4 millones de personas actualmente diagnosticadas y evitar la p\u00e9rdida de un total de 5.8 millones de a\u00f1os de vida ajustados por discapacidad (AVAD) para 2028, lo que tambi\u00e9n impulsar\u00eda el desarrollo de mejores tratamientos.<\/p><\/div><\/div>\n<\/div>\n<\/div><\/div>\n\n\n\n<div class=\"wp-block-uagb-container uagb-block-e40412f9 alignfull uagb-is-root-container\"><div class=\"uagb-container-inner-blocks-wrap\">\n<div class=\"wp-block-uagb-info-box uagb-block-eca68ba6 uagb-infobox__content-wrap  uagb-infobox-icon-above-title uagb-infobox-image-valign-top\"><div class=\"uagb-ifb-content\"><div class=\"uagb-ifb-title-wrap\"><h2 class=\"uagb-ifb-title\"><strong>Una iniciativa innovadora para el diagn\u00f3stico temprano y la mejora del manejo de la enfermedad de Parkinson<\/strong><\/h2><\/div><p class=\"uagb-ifb-desc\">La enfermedad de Parkinson (EP) representa un desaf\u00edo importante debido a su naturaleza neurodegenerativa progresiva, que afecta a m\u00faltiples sistemas del cuerpo y a m\u00e1s de 1,2 millones de europeos. A pesar de d\u00e9cadas de investigaci\u00f3n, nuestra comprensi\u00f3n sobre el inicio y la progresi\u00f3n de la enfermedad sigue siendo limitada, dificultando el diagn\u00f3stico temprano y el tratamiento eficaz.<br>Los avances recientes en la fisiopatolog\u00eda del Parkinson ofrecen esperanza para ralentizar la progresi\u00f3n de la enfermedad; sin embargo, revertir la degeneraci\u00f3n celular sigue siendo inalcanzable. Esto subraya la urgente necesidad de herramientas diagn\u00f3sticas innovadoras que detecten el p\u00e1rkinson en sus etapas m\u00e1s tempranas. Actualmente, el diagn\u00f3stico se basa en escalas subjetivas, como Hoehn y Yahr, o en costosas tecnolog\u00edas de imagen, sin biomarcadores validados disponibles para uso cl\u00ednico generalizado.<br>V\u03b1MPiRE busca revolucionar el diagn\u00f3stico del p\u00e1rkinson mediante un estudio longitudinal de casos y controles emparejados que involucra a 600 pacientes con p\u00e1rkinson y 600 participantes sin p\u00e1rkinson. Investigando isoformas de \u03b1-Sinucle\u00edna y biomarcadores relacionados en ves\u00edculas extracelulares derivadas de neuronas (NDEVs), nuestro proyecto tiene como objetivo desarrollar y validar una prueba diagn\u00f3stica in vitro (IVD) capaz de:<\/p><\/div><\/div>\n\n\n\n<ul class=\"wp-block-list\">\n<li>Detectar el p\u00e1rkinson en sus etapas m\u00e1s tempranas, cuando las intervenciones pueden tener el mayor impacto.<\/li>\n\n\n\n<li>Estimar el pron\u00f3stico de la enfermedad y monitorizar su progresi\u00f3n.<\/li>\n\n\n\n<li>Guiar las estrategias de tratamiento basadas en perfiles de enfermedad individualizados.<\/li>\n<\/ul>\n\n\n\n<div class=\"wp-block-uagb-container uagb-block-c3af8e7b\">\n<div class=\"wp-block-uagb-container uagb-block-1f437655\">\n<div class=\"wp-block-uagb-advanced-heading uagb-block-0c02a7b6\"><h3 class=\"uagb-heading-text\"><strong>Explorando el potencial de la \u03b1-Sinucle\u00edna y las NDEVs<\/strong><\/h3><p class=\"uagb-desc-text\">La acumulaci\u00f3n de prote\u00ednas \u03b1-sinucle\u00edna (\u03b1-Syn) mal plegadas es una caracter\u00edstica distintiva de la patolog\u00eda del p\u00e1rkinson y una v\u00eda prometedora para la innovaci\u00f3n diagn\u00f3stica.
